(lately),
Monday i spent pretty much all day at uni... started at 8am, finished at around 2:30ish...
Tuesday, woke up, cleaned, spent most of morning and half of afternoon driving... got to the lab and tried to catch up on lab work,
Wednesday, had to finish lab work left unfinished from tuesday... went to lectures and then went back to lab... stayed at uni studying till 6:30pm... went to gees and hung out till way too late....
Thursday: Now, obviously, went to lecture, coverslipped my slides ... went to PASS meeting...
and must study for BIOL3003 mid sem exam tomorrow (25%)

Now for more interesting stuff about me...
Been going okay apart from being perpentually tired which isn't very fun. Also been trying to spend the time i can with Gee, but sometimes i dont know whether he wants me to spend that much time with him (even though it might only be 2 or 3 days a week)...
I suppose its a good thing that we arent spending all that much time together then we wont get as bored... but i dont really like what it may say about our relationship in general. I guess i am thinking too much into it as always.

Hmm.. Parents? Well they arent divorced yet, but might be soon. My dad is back from poland and got himself all new clothes and toys and brought us back a few things from my grandma on mums side, but nothing from him. He got a new phone which i haven't even seen yet.. and gave his (new) phone away to someone in poland instead of giving it to my mum who has had her phone for years and really needs a new one as her battery keeps dying.

Am kinda stressing out about school... not the fact that its hard, but more the fact that i am finding it really difficult to sit down and actually get to do my work... I am not even working all that much, and i think my manager doesnt like me which makes work a bit more difficult, plus many people arent doing their job which makes my job more difficult.

Anyway, i am going to procrastinate in another way...
Ciaoz,

Ania

<msn:type>blogentry</msn:type><live:type>blogentry</live:type><live:typelabel>Blog entry</live:typelabel>

<dcterms:modified>2007-08-30T03:21:28Z</dcterms:modified>

An awesome piece of prose for an awesome night.

Hi guys,
Realise i havent blogged in quite a while yet again, however theres a rather awesome reason for my blogging today, my friend Andrew wrote the awesomest spiel about a party i went to on Sunday.
Full kudos and so much more to Andy!! He is going to make a great reporter.

==

==

Prohibition Party Laid Down by Law.

TEAGUE CITY: Flouting of prohibition was in full swing last night in
the usually quiet suburbs, as local politician and well-known
underworld figure Sam Law threw a swinging party to celebrate the 20th anniversary of his birth.

Resplendent in full flapper costume, stunning women from around the metropolis attended, escorted by dapper gentlemen in the finest attire of the day, including fedora hats, suspenders and that gangster favorite, firearms.

High-lights of the evening included high stakes poker games, various
gun duels and the constant salvos of flashbulbs as the party-goers
embraced "photo-graphy".

Several infamous and notorious characters of ill repute were in
attendance at the decadent affair including Christina the Gamblerina
and her male consort, Max "the Greek" Capone and noted physicist Simon
Bohr.

Also in attendance were world famous dancers Alice and Tyanna,
erotic novelist Mai Su, gentleman gangster Shanghai Jack, the
mysterious Raj Jee and his new flame-haired flame Ania.

Mysterious white-clad late comer Meg added some flapper grace and
class to the event, stirring up mischief and breaking hearts,
particularly Lael's! …And Ania's. …And Su's.

With his ravishing girlfriend Jess by his side, Sam was presented
with thousands of gifts, many of them from the mysterious east. Among
the booty were a shiny new watch, new decks of cards and four hundred
thousand pieces of drinking paraphernalia.

With the gambling, monopoly and suprisingly tame drinking games
raging into the early hours of the morning, junk food was consumed,
innocence smothered and foursomes formed.

There was no word today on the condition of the revellers, but hopes
are held they will be back to their usual level of mischief come
Tuesday.

There was also no comment from those who did not attend out of fear, laziness or crushing workloads.

Interesting Day

I woke up this morning to the sounds of my alarm 'hissing'. Hissing is what your radio does when theres no channel in that frequency and you have the radio volume low. That was my alarm, it decided to wake me up by trying to play a radio station and failing miserably. So i woke up 30 minutes later then i wanted. But thats okay, i've learnt that sleep is good.

So today, i spent about 2 hrs cleaning because we were having a house inspection between 10am - 2pm. I was ready at 10am, just for the inspection. They didnt come till 12:30pm so all that time i was doing absolutely nothing so i would create the least mess possible.. I didnt even eat because i didnt want to mess anything up!

And then.. i bumed around for a few hours, Dad came home with a new car.. a toyota prada, i think i am going to nickname it godzilla, because its from Japan and its massive!

Then i did some tutoring -- I love tutoring, gives me such a high.. its a good thing i've found a job where people actually listen to me!! I am going to love lecturing :P.

And then. i came home. had something to ate (because i was starving), watched the end of SVU and then Julian called me from Sydney and we had a nice long chat!!

So my day, which started on a rather crappy note, is ending amazingly well!!
And tomorrow is going to be tres fun!!

Can't wait!! *giggles like a lil catholic schoolgirl* (I dont think the catholic-ness has anything to do with it.. but i just put it in).

Oh and i got some new songs, J-T, Gia Farrell and the new unreleased FOB!! Awesomeness.

Oh and William Joseph is AMAZING! LISTEN TO IT!

Love lots,

Ania

"Noli me tangere, for Caesar's I am,
And wild for to hold, though I seem tame."

Sir Thomas Wyatt - Whoso list to Hunt

But they are fantastic!!

So, uni semester is coming up fast.. I've been pretty bored at home.. been to the gym everyday this week! I really do love exercise even if it tries to kill me... but I beat it everytime mwahaha... I am trying to get into the habit of exercising so I will want to go.. right now I go because I would die of boredom and I need to be kept occupied doing SOMETHING!.

Tomorrow I think I might clean my car.. it needs it alot. And then I shall go to the gym and do bodypump and hope it doesn't kill me like it did last time.. although it was the first time I had done bodypump in 2 years. I guess I deserved all I got!

Oh.. and I cut my hair! My mum hates it, I am starting to like it, its alot more manageable and easy but I do miss the long hair.. Oh well.. it will take about 2 years to grow back to how long it was.. shall be interesting.

I've also done my timetable and it is as follows:
Monday: 9am - virology lecture (1 hr)
10am - molec cell biol lecture (1 hr)
11am - dev. neurobiol lecture (1 hr)
7pm - tutoring cameron (maths) 1 hr

Tuesday: 8am - molec cell biol lecture (1 hr)
10am - genomics lecture (1 hr)
11:30am - tutoring amanda (statistics) ... not sure if this will go ahead, i dont feel competent tutoring her.. completely different way of doing stats then we did.
2pm - molec cell biol prac (2 hrs)

Wednesday: 8am - virology lecture (1 hr)
10am - virology prac (3 hrs)
5pm - tutoring caitlin (maths) 1 hr
6:30pm - tutoring cameron (maths) 1 hr

Thursday: 10am - genomics lecture (1 hr)
12pm - virology lecture (1 hr)
1pm - dev. neurobiol lecture (1 hr)
2pm - genomics prac (2 hrs)
6:30pm - tutoring vlad (maths/chem) 2 hrs

Friday: 10am - molec cell biol lecture (1 hr)
11am - dev neuro biol lecture (1 hr)

So my timetable isnt that bad..

i have 5 hours tutoring (not including Amanda), 5.5 hours mater work a week (really 5.5 x 3 a month.. we get a week 'off')

if i get a biol student i might take them on too, but only if.. otherwise i probably wont take on any more students..
and i get tuesdays and fridays all to myself!! yay!!.. gym and study time..

I really want to study hard this year (i know i say that every year).. but this year i really mean it!!!

Anyway, should go!!
Thankyou all for helping me hit 10,000 accesses!! I've been single-stranded too long! Lonely ATGCATG would like to pair up
with congenial TACGTAC.

Menage a trois! Ligand seeks two receptors into binding and mutual
phosphorylation. Let's get together and transduce some signals.

Some dates have called me a promotor. Others have referred to me as a real
operator. Personally, I think I'm just a cute piece of DNA who is still
looking for that special transcription factor to help me unwind.

Highly sensitive, orally active small molecule seeks stable well-structured
receptor who knows size isn't everything.

There must be a rational way to meet a date! I'm tired of hanging out in
those molecular diversity bars, hoping to randomly bump into the right
peptide. I want a molecule that will fit right into my active site and
really turn me on. I'll send you my crystal structure if you send me
yours!

Gene therapy graduate. After years of producing nothing but gibberish, I've
shed my exons and am ready to express my introns. All I need is a cute
vector to introduce me to the right host.

My RNA, I'm sorry I misread your UAAUAAUAA and inserted three tyrosines
when you repeatedly asked me to stop. Something got lost in the
translation. Please forgive me.

Naked DNA with sticky ends seeks kanamycin-resistant plasmid. EcoR1 sites
preferred.

Uninhibited virus seeks reason to make me shed my coat protein.

This very selective oliogonucleotide has been probing for just the right
target for long term hybridization.

Mature cell seeks same who still enjoys cycling and won't go apoptotic on
me. Let's fight senescence together!

I'm a prolific progenitor with great potential for growth and self-renewal.
Call me if you're a potent hematopoietic factor who still believes in
endless nights of colony stimulation.

I don't always express myself on the surface, but I'm looking for a signal
that you appreciate my complexity. Send me the right message that will
penetrate my membranes, turn on my protein expression and release my
potential energy.

A new dramatic series set in the hustle bustle world of high fashion
protein synthesis It's ...

                                 ROUGH ER

See the drama of these hot young ribosomes as they fight to save proteins,
surrounded by the harsh and cruel reducing environment of the big
cytoplasm.

Watch as the mutilated remains of spliced and modified RNA come into the ER
to be transformed into hardy new proteins. Ready to take on anything the
extracellular environment can throw at them, as they go for the Golgi.

Stay tuned to your TV listings for where to tune for

No-one knows exactly what to do, do you go to work? (if your lucky enough to have work!), do you go shopping? (even though you already have everything you want, and anything you do want, you know you won't have the time to enjoy --- like a Wii!!!!). Do you play with all your xmas stuff? (that is if you got toys..), Do you read a book? (i need a book)... Do you just idlely waste your time like you have the last few days under the pretense of 'xmas holiday'?

How about we do some homework?
Find out about different cultures, i am sure not all of earths billions of people dont celebrate xmas, actually i would be surprised if half of them did. -- not even everyone celebrates 'our' new year, some people dont follow our gregorian calender.
Actually, did you know, that until 1990's the University of Helsinki had a monopoly on the sale of calendars in Finland? I didnt until wikipedia told me.

So maybe, instead of trying to grab all the cheap stuff from Myer, we should be thinking about people who do things a little differently from us?

Have a good new year everyone.

-- Ania

<msn:type>blogentry</msn:type><live:type>blogentry</live:type><live:typelabel>Blog entry</live:typelabel>

<dcterms:modified>2006-12-28T02:36:47Z</dcterms:modified>

Pink rocks.

I know I haven't blogged for a very long time, since november... and its now nearly end of december, but i have a shocking headache so I am not sure what kind of entry this will be. Most likely all over the place per usual :P.

Haven't been doing too much, just working on my summer semester project, working in general at mater and tuition (have done my last tuition there!!! yay -- I'm free!!!).
Looking for work at the moment, just anywhere, will probably end up in retail or something, (because ofcourse no-one employs science students to science related jobs *gasp!*). The mater job is really screwing me up, the lifts and the buzzing of the computers in the office (they are really old computers) gives me vertigo and makes me sick. Plus i have a moral thing against the actual job, even if it does pay well.. i feel like i am selling my soul.

So, i dont really know whats happening, i havent been paid yet for the research project (its supposed to come in instalments)...
And christmas shopping is draining my funds.. :P

Anyway,

I hope everyone has a great christmas and new year!!

Love,

Ania

<msn:type>blogentry</msn:type><live:type>blogentry</live:type><live:typelabel>Blog entry</live:typelabel>

<dcterms:modified>2006-12-18T02:24:09Z</dcterms:modified>

Tired

I am really really really tired :(.

If anyone wants to do something on sunday or sometime next week, message me and i'll organise something..
I am working tomorrow from 3:30-7:30 and friday from 5-6, and all day saturday (but i might go out sat night..!!!)

Ciao
Ania.

*collapses*

<msn:type>blogentry</msn:type><live:type>blogentry</live:type><live:typelabel>Blog entry</live:typelabel>

<dcterms:modified>2006-11-15T11:09:20Z</dcterms:modified>

Its all over

Well thats it, a whole semesters worth of work assessed in just a few very short hours that i usually cut short early by graciously walking out when my head feels like imploding.
Its hard to imagine why examiners thought up of this method of testing knowledge.. why confine hours of work into very short space where only a small fraction is covered even though theres a deception that everything MAY be on it, thus you need to know everything.
It feels like a bit of a disappointment to everything else that was covered... where is the respect nowadays? Crammed into 100 marks while assessments assess something completely foreign to what the curriculum actually encompassess.

I am getting some sleep ^.~ Ironically, i have a long day tomorrow...

Night,

P.S.

THE POTATO STORY

Well, Girl Potato and Boy Potato had eyes for each other,
and finally they got married, and had a little sweet potato,
which they called 'Yam.'

Of course, they wanted the best for Yam.

When it was time, they told her about the facts of life.

They warned her about going out and getting half-baked,
so she wouldn't get accidentally mashed,
and get a bad name for herself like 'Hot Potato,'
and end up with a bunch of Tater Tots.

Yam said not to worry, no Spud would get her into
the sack and make a rotten potato out of her!

But, on the other hand, she wouldn't stay home
and become a Couch Potato either.
She would get plenty of exercise
so as not to be skinny like her Shoestring cousins.

When she went off to Europe,
Mr. and Mrs. Potato told Yam
to watch out for the hard-boiled guys from Ireland.
And the greasy guys from France
called the French Fries.
And, when she went out west,
to watch out for the Indians
so she wouldn't get scalloped.

Yam said she would stay on
the straight and narrow and wouldn't associate
with those high class Yukon Golds,
or the ones from the other side of the tracks
who advertise their trade

on all the trucks that say, 'Frito Lay.'

Mr. and Mrs. Potato sent Yam to Idaho P.U.
(that's Potato University)
so that, when she graduated,
she'd really be in the Chips.

But in spite of all they did for her,
one day Yam came home and announced
she was going to marry Eddie McGuire.

Eddie McGuire!

Mr. and Mrs. Potato were very upset.
They told Yam she couldn't
possibly marry Eddie McGuire
because he's just . . .

Are you ready for this?

Are you sure?

OK! Here it is!

Hey!

Going to add a few photos to this post as penace for not putting photos up the other day!

Oh and to all my brissie friends -- expect an email soon about a certain event during midsemester break!!! Don't scull coffee... just dont.. its a REALLY bad idea... unless you want to look like a dog trying to get peanut butter off their tongue.
2. Sleep is important!
3. If you wake up in your sleep screaming in pain... something is definitely wrong!
4. oh and food IS good for you.. stops you from shaking ^.^

So... you may be asking what busy ania is up to today...
WELL
i have lectures from 9am - 11am (cell and dev and biochem... histol was cancelled)
Then i am going to IMB lecture on telomere biology, then i am going to study my slide for histology till about 2pm.
THEN drive to Milton and pick up $100 worth of polish meat... then pick my dad up from the city.. take the meat and my dad back home... then drive my dad to Manly.
(yes i am playing taxi today...)

So... busy :)

Anyway.. have a great day.. pictures will be added when i get home.. whenever that will be ^.~

Love,

--Ania

<msn:type>blogentry</msn:type><live:type>blogentry</live:type><live:typelabel>Blog entry</live:typelabel>

<dcterms:modified>2006-09-14T22:25:38Z</dcterms:modified>

Mater talk

Today when i was working at the Mater and with a patient ['How many days do you think you are in for?? Oh.. okay you realise it finishes 4pm the NEXT... its cheaper if...'] when i overhead the couple in the next bed (this was at the mothers maternity hospital). The woman was saying how good the meal had been, because she didnt have to cook, and the husband replied with 'and you wont have to cook' and she made a 'what?' sound, and he explained that she wasnt cooking for two weeks when she got home because he didnt want her to do anything strenous. It was so sweet. (afterwards they launched into a conversation about whether or not the baby could hear them).

What was so significant about it (and why i wanted to blog it) was because it portrays the kind of relationship i like to think i have with Mitch. We both care immensely for each other, even if we dont completely or even half-ly understand each other, and we just do the little things that help each other get through the day... like a sexy mms, or just a funny pick up line.

... Yes, you can all go puke now.

I am off to bed!!

'Nite!!!

[I miss you like a fat kid misses a cupcake... thats a WHOLE lot...] {In a SMS from Mitch :P}

Love,

Ania

P.S. if your wondering about the picture, it was just cute... and its holding a cupcake!

<msn:type>blogentry</msn:type><live:type>blogentry</live:type><live:typelabel>Blog entry</live:typelabel>

<dcterms:modified>2006-08-06T13:13:38Z</dcterms:modified>

Moulds, Drugs and other elastic things

Wow, spaces has changed... *pretty* ... sorry about the title, it sounded cool!
I was actually thinking of going to sleep, but then i realised i am too hyped up (i ate like 3 tim tams before) to sleep.. so here i am.
I just watched a pretty cool documentry on SBS about the discovery and manufacture of penicillin. I think i did a spiel about it on this blog, so i am not going to do it again, but to cut a long story short, Fleming, the man we all seem to credit with making penicillin a household name is actually just the man who discovered the mould and its bacteria-eating abilities. (Which is pretty amazing in its self... but..) Fleming didnt do the hard work isolating the active ingredient from the mould (what we now call penicillan), he didnt do any of the work testing it, or figuring out a way to purify sufficient amounts of it in high concentrations, all done during WW2 where funding for a unknown scientific compound was non existant and they had to use the cambridge chemistry department as a mould factory. It took them 2 years to isolate a small amount of just pencillian.. and then the tests.. first on mice and then clinical trials, and all this was done by australian scientist Florey and his chemist, Chain and pharmist/chemist/engineer extraordinare Heartly.

Unfortunately only Fleming, Florey and Chain got the Nobel prize (although Heartly was the first person to be awarded an honoury doctorate from Cambridge university).

Now, where you ask, does fleming come into this, well he realised the potential of penicillian when the boys from cambridge began to do test trials, and he pretty much stole the publicity and drove all the credit in his directions.. when all he had done was been careless with a plate of bacteria, left them on a plate open overnight where spores of the mould dropped on them and ta-da.. not really very brilliant when it was all an accident. This was at a time where patents were considered immoral and cash sucking... if they had patented it, this problem wouldnt have arisen.
To bring things back to current events, Ian Frazer of UQ is currently having problems with his right to fame with his viral cervical cancer drug because of patent irregularities. (there are actually 2 other universities claiming to have discovered the use of virus-like particles (VLPs). [If you were wondering who the other two unis were, they are the University of Rochester and Georgetown University]. Fortunately, the drug companies who have deals with these labs, Merck and GlaxoSmithKline (GSK), have reached a side-stepped agreement so that any disputes between patents will not affect the manufacturing of the drug which will potentially save millions of women. It has already been approved in the USA by the FDA and is up for approval in Australia.

So, to wrap things up.. because the chocolate seems to be wearing off.. Fleming didnt have much to do with the actually making penicillin useful to us, he just discovered that it may be of use... and left it. The real work was done by Florey, Chain and Heartly.

The pictures attached are:
1. Pencillin mould eating a colony of bacteria (the white-blue squiggles are the bacteria, large white round thing on the bottom is the mould, you can see a clear boundry between the two where the penicillin has degraded the bacteria).
2. The chemical structure of penicillin (there isnt just one penicillin molecule, there is a whole class of them).
3. The japanese pop-band 'Penicillin'
4. Yes i realise there was no picture... the Americans apparently named a crater on the moon after Fleming, i wanted to see if i could find a picture of it, but i couldn't.

<msn:type>blogentry</msn:type><live:type>blogentry</live:type><live:typelabel>Blog entry</live:typelabel>

<dcterms:modified>2006-08-03T12:17:51Z</dcterms:modified>

NEW RESOLUTION

I am making a new *semester* resolution.. (its only a new semester resolution because thats pretty much the most recent event i can bung this onto, its not exactly the new year...)

So, all of you must be wondering what this resolution actually is! And i shall tell you..

RIGHT...

...

..

..

.

NOW!
My new resolution is to be healthier and take better care of myself. I have come to the realisation (after skipping a meal today and feeling very woozy and having the sensation that i wasnt actually standing on the ground at work) that being even a little unhealthy.. like missing meals or a few hours of sleep is really not doing me any good. I suppose getting up everyday but sundays at 6am and only getting about 6hrs sleep everynight isnt a good thing either.

I suppose i should list all the bad things i do, might make me more aware of the things i need to change:

1. Constantly sleeping too little

2. Skipping meals

3. Eating unhealthy (living off museli bars, chocolate, chips etc)

4. Not exercing enough (causes an increase in tension from stress)

5. Bottling up stress and negative emotions

6. Not having any 'breathers'

7. Stressing too much

8. Being lazy :P

ANyway, i cant think of anything more.. if you think of something, just comment :P

Hopefully i can fix a few of these.. *fingers crossed*

--Ania

Originially from a email sent to Richard :P

Hmm.. had a biochemistry lecture (Actually called cellular biochemistry of health and disease... but biochem is so much easier to say) was fairly interesting, cheaper foods use methods to make polyunsaturated fats into polysaturated fats by hydrolysis, and a byproduct of this is trans fatty acids, which are really bad for you (as opposed to good natrually occurring cis fatty acids ... difference between cis and trans is how the hydrogen groups allign themselves on either side of the hydrocarbon chain). They are bad for you because they increase the amount of low density lipoprotein (LDL) cholesterol which is bad for you.. and decrease the amount of high density lipoprotein (HDL) cholesterol which is actually good for you!!! Consumption of lots of trans fatty acids is a proven factor in coronary diseases!! -- SO dont eat cheap foods!! (i.e. Maccas, 'Black & Decker' Homebrand etc..)

Fingers crossed!!!

The main character, Hayley, everything she did, i could see it as either a physical juxtaposition i have done or a metaphor for something i have done.. So it was rather painful to see her do such things to someone else and what it feels like for the recipient.
It movie carries a very strong message and i think its a very powerful movie.
I wont say any more about it and leave you to make your own decisions about it, but do go see it.

<msn:type>blogentry</msn:type><live:type>blogentry</live:type><live:typelabel>Blog entry</live:typelabel>

<dcterms:modified>2006-07-19T08:02:10Z</dcterms:modified>

101 Turn offs

~ Written by Jess and Ania, when Ania was in perth :P

Why didnt i study?? :P Oh well, i dont need it, and i didnt apply for a supplementry exam, because i might need it for subject i actually need which i collapsed in an exam :P (it happens!)

I am a little worried about next semester, i am going to be fairly busy here is my timetable so far:

Monday
9am - BIOL2012 lecture
10am - BIOL2008 lecture
12pm - BIOM2034 lecture
2pm - 4pm BIOM2034 prac
5:30pm onwards - possibly tutoring at clayfield

Tuesday
8am - BIOL2008 lecture
9am - BIOM2034 lecture
11am - BIOL2007 lecture
2pm - 5pm BIOL2008 prac
6pm-7pm tutoring at bulimba

Wednesday
8am - BIOL2012 lecture
10am-1pm BIOL2008 prac
1pm - BIOL2008 lecture
2pm - 5pm BIOL2012 prac

Thursday
11am - BIOL2008 lecture
12pm - BIOl2012 lecture
2:30pm - 6pm tutoring at Bulimba

Friday
8am - BIOM2034 lecture
10am - BIOL2008 lecture
3pm - 7pm - tutoring at carindale

Saturday
8-9 tutoring at bulimba

Interchangable sat/sun
10:30am -1/5pm-8pm (split shift) - working at Mater hospital for entertainment network

And this timetable does not include the stat pass i am doing (which is confirmed that i did get through).

I am a little worried as to when i can see mitch and keep up with all my schoolwork at the same time.. there is a reason i am going to uni.. and i really want to do well. Maybe i have bitten off more then i can chew?
I suppose the real crunch time will come when semester starts, i can always drop things.
Oh and another note about the timetable, i will probably be getting to uni everyday before 8am because of the shitty dutton park parking!

Talking about Mitch, everything is going fairly well, we had a nice chat today to see what direction we are going in and what we can fix. Hes so sexy! :P Sunday is our 3 month anniversary but i dont know when i will see him next :(

Hmm... what else.. i made a new cd for my car.. yayness..
oh and i watched lord of war.. i really liked it, except i couldnt come to grips with the emotionlessness of the main character, even though it was exceptional acting. Feel free to think differently!

I am still yet to see pirates of the carribean 2, so if anyone is organising something, count me in! or maybe i can organise something, now theres a thought! :P

Anyway, i think i have written enough! its going to be too long! Dont forget to check out the whitsunday pictures, i know they take forever to load, but there are some funny ones in there!

Love,
Ania.

<msn:type>blogentry</msn:type><live:type>blogentry</live:type><live:typelabel>Blog entry</live:typelabel>

<dcterms:modified>2006-07-09T19:43:26Z</dcterms:modified>

Whitsunday Holiday

Hey,

Just a quick post to say that i've put up selected photos from my families and I holiday from the whitsundays so browse through.. theres a fair few so it may take a while to load (77 in total i think).

It was fun! the weather was a little off though and i was sea sick the first day (vomiting is not fun, i could never be bulimic), the seas were too rough to make the trip from Butterfly Bay to Whitehaven beach, so that kinda sucked, but we got to see hamiliton island, so that was cool!!

Had romantic fish and chips with Mitchie on the harbour :P

Hopefully i can update some more later, but thats about it for now!

I want to study you!!). Even though we are always in contact with our little microbial partners, many people who dont undergo the torture of a science degree (with a slight focus in microbiology) dont actually know much about them.

So i am here to slight enlighten you.

*Throughout my spiel, please remember the central dogma of genetics:

DNA --(transcription)--> RNA --(translation)--> Protein

Also, prokaryotic cells (which include bacteria and viruses, not humans, we are eukaryotes) do not undergo post translational modification, such as splicing of introns and the joining of extrons such as what happens in eukaryotic cells. However.. viruses are a little tricky and do undergo some modifications... but more on this later!

First off we shall explore the arch enemies of bacteria and viruses- antibiotics.

When the word antibiotics comes to mind, most people think of penicillin and its famous discoverer Fleming.

The first substance to have selective toxicity on bacteria (i.e. would usually specifically target bacteria, but in early cases could have also affected other things) was Salvarsan which was developed in the early 1900's by Ehrlich as a effective treatment against syphillius (a STD which usually produced fatalities) however it had rather toxic side effects.

Lets now focus more on the superstar of antibiotics - penicillian. Discovered by fleming in 1929, an often misconception people have is that Fleming actually had something to do with it becoming a widespread antibiotic that helped millions of people. When in fact, (and i didnt know this either until the lecture), Fleming accidently left a petri dish of bacteria on a bench which were contamined by a mould, overnight, and when he came back in the morning, he found that the contamination had inhibited and killed the growth of the bacteria on his plates. He named the mould 'penicillian' after the genus name of the mould (penicillian penicillan). This is where Fleming stopped.

It was little known Florey who actually developed a process to manufacture large scale production of penicillian from the late 1930's to early 1940s. He was also the person who demonstrated that it was effective in treating infectious disease.

Fleming, Florey and (a partner) Chain won the Nobel Prize in 1945.

Now as many people know (or should know) soon after penicillian was introduced into the world, there were cases of people being resistant to the drug. Doctors and scientists were stumped.

What they didnt connect was that penicilian was a natural substance, a mould which grew in the ground. Why wouldnt diseases which competed with penicillian for nutrients have created their own resistance to it?

And this is just what happened - A few years after an antibiotic was released, the disease had already evolved to be resistant to it.

Cells are smart... they evolved to create enzymes which could chop up antibiotics to inactivate them or some pumped the antibiotic out of the cell so it couldnt build up in large concentrations which would kill the cell. Or perhaps they can degrade antibiotics?

There is evidence viruses and bacteria not only rapidly evolve to new antibiotics but have already possibly (even decades before) have evolved for these antibiotics..

The main problem with antibiotics is the reliance of natrual molecules as antibiotics because the diseases have probably already had interacts with them.

Bacteria are also very sneaky microbes. They have these extra circular molecules called plasmids, which can hold self replicating genetic information (apart from the cellular chromosomes of the bacteria). Now these plasmids are key to antibiotic resistance because this is the site where antibiotic resistant genes are most likely to occur. As a bonus, the bacteria can directly uptake these resistance genes (as DNA ofcourse) from the environment, however this can only happen between the same species of bacteria.

Bacteria can also 'transduct' or transfer a copy of its DNA to another bacteria of the same species.. this is called horizontal gene transfer.

However the most kinky and fascinating mechanism is conjugation, or bacterial sex as it is understood in the microbiology world.

Conjugation is where the plasmid itself mediates the transfer (controlled and initiated by the tra genes on the plasmid) of a copy of its own plasmid to another bacterial cell.. regardless of what type, genus, species it is.

It does this by extending a thin 'feeler' or sex pilius to a nearby bacterial cell and pulls it in closer to it, then extending the pillius to a bridge which the plasmid genetic information can travel down and assemble back into its plasmid circular structure at the recipient cell. This way, a possibly completely foriegn cell can trade genetic information ... and this information can be on anything... commonly antibiotic resistance but also mechanisms for combating possible obstacles within the environment such as a nutrient shortage or temperature.

So.. we have bacteria who already have dealed with our antibiotics, and have mechanisms which can very quickly adapt or trade genes for adopting resistance to bacteria. We are screwed.

Bacteria have also created integrons which combine a single promoter for a whole bunch of genes on a plasmid so that it takes less time and energy to create or move (to another plasmid) antibiotic resistance genes.

IN ADDITION (yes theres more) viruses and bacteria have figured out that there are features about them (such as surface expressed virulence factors... think tentacles and such) which induce an immune response, so they can randomly switch them off to not induce a response and then randomly switch them back on again.. cool huh? Its called phase variation.

There are lots of other things.. but i am really tired now.. and hungry.. so i might continue later, i might not, either way i hope you have gained some sort of insight into the microscopic world living in, on and around us.

P.S. Panic! At the Disco is an awesome band!! Check out their song - 'lying is the most fun a girl can have with her clothes on!'

Ciao!

--Ania

Today shouldnt be too bad, i've nearly finished uni (just one genetics
lecture left, and i like the lecturer..) and then i have several hrs of
work (i can finally eat!! i am starving!!)
*Listening to within temptation from my *new* USB Drive.. 1gb.. oh yeah.. *

Hmm.. I felt rather strange last night, it was the first time in
a few years and i felt particularly bad .. (the people who have known
me for a few years may or may not know what i am talking about). I
would like to publically (as publically as I can) apologise to Mitch..
for freaking him out last night.. sorry.. i didnt expect you to
call me.. and i am pretty sure i sounded alot worse then i was. I guess
everything just got to me and that was one factor that was continously
trying to bite me. (alot of things try to bite me.. i have 2 large
bites from something.. thinking its a sand fly.. but why cant they just
leave me alone???)
Um, so yes sorry.. i will explain to you if you really want to know.
but its just emotional baggage shit that doesnt usually matter... Only
once in a while during my crazy periods :P

Anyway.. i should go coz i have nothing else to say!!

* Ciao *

--Ania

P.S I am pretty sure 'ciao' can be used as hello and goodbye, my friend matt seems to disagree with me.. ?? Any input?
Its 8:19am, i am sitting in iLC 2 (which is pretty cold compared to iLC 4), and i am contemplating.
What are you contemplating you may ask? --Well i am not sure yet, i suppose i am contemplating what to contemplate.
As you may have noticed my brain is currently still asleep, even though i have dosed it with strong caffeine to jump start it, however i doubt that will last until my 10am physical chemistry lecture, and as usual my head will want to draw demented animals all over my lecture notes.
I have noticed that my language has quite rapidly degraded into a more common and less complex vocabulary, so i am going to try and force it back up to the level at which it rested when i graduated high school -- i cant let all those years being prepped by ms. Kav go to waste!! It was so much fun!

Does anyone know how to calculate the temperature two solids are at equilibrium when given the phase transition and freezing point at a certain pressure??
Or the value of change of entropy for a reaction when given heat of vaporation, boiling point at a certain temperature and pressure?

Or perhaps calculating an equilibrium constant a reaction of NO at 1000K when given the standard free energy ?

Hmm...

Oh! My laptop is probably getting sent over to Dell on monday so from then on i will have restricted access to computers -- ie. email, messenger etc, so please bear with me.. i will be online at uni and stuff to do assessments but it wont be as much as i usually am.

Gracie!!

If it went on much longer the mechanism which pushes it open would probably break from their being something pushing against its force.
So i am happy :)
AnD i am seeing my genetics co-ordinator about my mid sem result (and hes going to help me figure out how to answer MCQs efficiently), i also want to talk to him about possible labs to volunteer at on the holidays to get some proper lab experience.

Still 15 minutes before i even think of chemistry.. hopefully i will pass my chem CMT (computer managed test) which i need to submit today... i did kinda work on it *sheepish grin* not sure why i even took chem1013 or biom2007 (physiology), both are pretty annoying.. although for physiology we now have all the cool lecturers!

I cant believe how well our calcium prac went yesterday for chem.. it was so awesome.. i finally got a column to work!! (although it was a resin column not a matrix column like in BIOC2012.. and we didnt completely set up the column, although i still had to neutralise charges and such.. took a while, nearly an hour because the column let the liquids flow down drop...by...drop... :P

we got 9/10.. we lost one mark for prelabs, but it was tricky!! (i swear!)
*wonders why she is listening to MiB...??)

*sings: where have all the good men gone... and where are all the gods?? wheres the street wise hercules to fight the rising odds... isnt there a white knight.. upon a fiery steed.. late at night i toss and i turn and i dream of what i neeeeed......... oooohhhhH!!! I need a hero... i'm holding out for a hero till the morning light!*
Hehe.. can anyone guess what movie that is from?!?

I love that sing.. has awesome instrumental!!! (just the reason why i love 'this is how a heart breaks' because theres an awesome violin part in the middle of it.. its so kickass!!*

Also like nightwish and apocolyptica for that reason although to be honest havent listened to them in ages.. might put one of their CDs in on the way home (currently have Cursive in there).

Its really cold..........
And i didnt go for a walk this morning (seriously did contemplate it though does that count?!) coz it was too damn cold and i can go for one this afternoon... before or after watching NCIS.

Yesterdays episode of House was also pretty interesting.. poor Chase.. will be interesting though.. Foreman as boss? And is house going to try and ruin him because he knows foreman may be potentially up to the job??

ANYWAY...
i might wander down to Hawken and see who has gathered for chem... need to go through my cmt with amanda, need her opinion on several things...

Ciaoz!!

(this turned out to be a longer posst then i thought... :P :P :P)

<msn:type>blogentry</msn:type><live:type>blogentry</live:type><live:typelabel>Blog entry</live:typelabel>

<dcterms:modified>2006-04-26T23:45:37Z</dcterms:modified>

Things are good!

Hi everyone,
I know last time i blogged, things werent going particularly well.. i
had just been rejected for a perfect position, i was having familial
issues, and was dreading going back to uni.

Well, this week has been alot better even though some very nasty things have happened.
I think it seems like the week is so good because i now have a very
awesome and gorgeous boyfriend who is very comforting and cuddly and i
just love spending time with him.
Anyway, enough about gooey stuff :P
I got 50% on my genetics mid sem, which i was pretty upset about, its
good that i am not a person who keeps things bottled up, i can just let
things go and hope for a better result next time, but i really tried
hard, and studied alot.
I also got 57% on biochem, which i can understand although i also studied that alot and was still fairly disappointed.
You can imagine the day i had... these results were released almost on the same day.
And the genetics lab i attended, was really a tutorial for going over
some data attained in the last lab, i didnt understand anything.. which
isnt good.

So i am considering dropping a few hours at work so i have more time to
study, because my grades are really suffering and i do want to do
really well (there goes the idea of getting a GPA of 6). So hopefully
with a few hours more time, i can be a little less stressed and
actually get some work done!!!! (Because at the moment i cant really
study because i am too tired and other distractions are too enticing...
that and because of my stress i cant seem to retain anything).

I have also applied for a job at a pizza hut call centre in moorooka.
Its alot closer to home then subway in the city (although subway is
close to Mitch but i dont think i would want to go over there after a
shift all smelly and sweaty and tired).

Oh, and i spilt orange juice on my laptop keyboard... but dont worry,
it still works, some of the keys are just hard to press.. and i have
accidental warrenty on my laptop, i still havent gotten through to Dell
yet, but i will do it when i get home.

I spent the whole day yesterday with Mitch and it was great. Not sure
if i will survive till Monday to see him again. Oh well. I'll have to
manage somehow.

Just waiting for people from genetics come to this building and hand
out the exam papers.... I am seeing the course co-ordinator about my
result because i got 0 for both multiple choice sections and i was sure
about some answers.

ANYWAY... i really should go do something.. like my CMT (which i have
to finish by tomorrow but didnt bring to uni today because i thought i
would be too busy... i didnt think i wouldnt have a physiology prac i
was all prepared for... its next week....grr)

Anyway.. I will leave you nice people to get on with whatever you were doing before you went.. oh look ania blogged again!!

Over and out
P.S SO going to the wolfmother concert!!!

<msn:type>blogentry</msn:type><live:type>blogentry</live:type><live:typelabel>Blog entry</live:typelabel>

<dcterms:modified>2006-04-26T02:14:34Z</dcterms:modified>

Easter with no eggs..:P

Hello my adoring little fans.. and yes i am joking.. your all probably taller then me.
.. and i am sure your not my fans. you just read this because your probably bored.

I got back from Noosa today.. yup.. spent Easter there with my family.. didnt really come back feeling all warm and fuzzy... as we all know, family dysfunctia is a virus that is fast spreading through the western (and probably southern, eastern and northern) worlds. As yet, there is no cure, but there is always hope. Always hope.

I was going to talk about something interesting.. like, the human genome, mRNA, ribosomes, chaperones, chaperonins, ubiquitin or proteasomes instead of my usual boring, mindless, argh!!! rant i usually give on here.

However, i dont know what i will say.. because i dont really feel like giving a science spiel, and i dont really want to rant on about how my job sucks and i cant seem to get any interviews for any sciency jobs -- lab assistants (i.e. rats), medical administration staff.. no-one.. it sucks. i have applied to over a dozen places.

Anyway, i think i will end this here..
Hope everyone had a happy easter weekend!!

Love,
Ania
<img sty
